Can Desktop Office Integration (DOI) be used to open Office applications(Word/Excel) that are stored in application server?

Not really, no, as it is dependent on the OLE capabilities of SAPGui. If you can give the presentation server access to the application server file system, then you can do it.

The presentation server is your pc, right? So your Basis people need to set up some way that you can access a folder on the app server, as though it was a network drive.

Talk to them, they'll know what to do.
