I need toimpliment this note and not claear about third step mention in the note.

1) Download the CSV file attached to this note. Do not open the file with Microsoft Excel, as this may garble its content.

2) Execute function module E2E_UPLOAD_TEMPLATE_TABLE in transaction SE37 and enter the path to the CSV, execute. This will update the template table defining which extractors are to be started.

3) Run the setup wizard for managed systems again. If the system is supported, the appropriate extractors will activated in the Extractor Framework and a controlling batch process will be created.

Can any one help me in understanding the step 3.

second thing is since this is manual implimentation of the note do still need to use SNOTE transaction and upload the note there?

It is always good the use SNOTE rather you do it manually. Please try the same with SNOTE.
