Charm means SAP's solution manager is involved https://blogs.sap.com/2009/11/27/the-complete-charm-solution/.

Manual request should be a request that is transported manually with SE01 / STMS.

It's a big shortcut, check the white book of SolMan. There are a lot of tools based on SolMan not related to Charm.

Charm is a tool installed in a Solman System.

  • it is used to follow the Change management (there are several tools behind, I put everything behind Charm to simplify)
  • it could be used to generate Transport Request for the SAP Systems.

In general, Charm also manage automatic Transport Of Copy (TOC for friends), gives you the ability to import Task without releasing your transport request (it create a copy of the request for a one shot).

Normaly, when you use Charm, you have to lock the TR creation on the system managed. But, if you do not have this, you could create a MANUAL REQUEST. (means TR created directly in the managed system without using Charm)

So, to know, if it is managed from Charm, you have to connect to Charm and use a report to search for this TR.

You could also check the naming of the TR: S <change_number>: <description of the change number> but people could easily simulate this.
