In above case /usr/sap/trans should share with additional instance or not?

Raj,

You must have /usr/sap/trans shared across all the application servers. An SAP system does have only one trans directory to setup TMS. This trans directory may or may notbe shared across all the two or three systems of the landscape.

How else you think a transport generated in one application server will be notified to the system? Had it been so, it would be a huge insocistency in data changes.

Hi Raj,

Do you mean an additional application server instance (aka dialog instance) as part of the same S/4HANA system? Or do you mean a separate application entirely, with its own System ID, running a different product? I think you mean the former, in which case Sumit's answer is correct: you must have just one \usr\sap\trans folder supporting the entire transport domain (DEV, QAS, PRD for the same application).

Cheers,
Matt
