A database I am looking after is experiencing performance problems. I think it could be contention for locks or/and hot rows.
How best would I evaluate this theory?

I recommend looking at Dan's whitepaper on performance analysis, and the whitepaper Ivan and I wrote on capacity planning (particularly the section on convoys and lock contention).
