B2B adapters enable the cross-company communication? or some kind of tunneling must be in place before hand?

B2B adapters are third party adapters which will be used in B2B Communication.
Ex: Seeburger, iWay Adapters....
When you are dealing with in your Organization levele or Network level means it is A2A Scenario
B2B Means the communication between two Organizations or parties
A2A - application 2 application:
this is a integration like let's say, you have 2 systems in your company: ERP & accounting system. And you want to send data from ERP to acconting system. This happens inside your company, so it is sol called A2A.
B2B - business 2 business
Let's say, there are 2 companies, and they have a different systems, which need to be integrated. For Ex: your client is using SAP R/3 and their customer is having some other technology. they want to integrate each Appications. this is B2B

Yes,you need extra settings/configurations for B2B scenarios.
These settings depends upon the adapter you would be using,for e.g if you want to use AS2 adapter,you need to have AS2 server in place,similarly to use FTP VAN you need to have mailbox setup on VAN.
In addition to above ,to Install external B2B adapters,you need settings on XI server as well

those kind of adapter usually allow you to connect with different applications natively
(so you send a message from XI and it's translated to the destinations applications message (it can use
non XI supported protocol (SFTP etc) ) for exmaple
if you want to use ARIBA then probably the application will be Ariba Value Chain Management

B2B Adapters are third party adapters. It is developed based on the industry standards. Like iWay,SeeBurger etc
Technical adapter are protocol specific (like JMS, JDBC, HTTP)
Business Adapter (B2B) are business specific. They work mostly with business objects then messages. Business adapter can use standard protocol. For example Rosettanet uses HTTP/HTTPS.
