Does a machine running an application referencing the SAPBusinessOneSDK.dll in order to create a transaction in SAP need to have the DI API installed on that machine?

Yes, the DI API has to be installed.
When the SBO Client is installat the corresponding version of the DI API is also installed.
Thanks for you answer!
What if the DI Server is used to create the transaction in SAP instead, is the DI API still required on the machine?
In that case the DI Api is only needed on the machine with the DI Server.
