Is it MANDATORY to enter condition type SKTO, WOTB & NAVS in cal schema.

No its not mandatory to enter the above mentioned condition types. Its your choice, if you want to add them or not in your calculation schema.

Why it will have impact on printing PO output. For PO printing, message records are set in MN04, and scripts are maintained through ABAP for PO print. Yes, if the conditions mentioned above are hard coded on PO print, it will impact on PO print, but if they are not hard coded to display these records, it will not impact.

It depends on the companies requirement what type of layout for PO print they want to choose.
