I am getting this error msg when trying to create an STO for moving Parts from a PO,  All other parts of SAP are working fine, This just started to happen one day and we are very confused as to why,

Looks like a Custom Solution.
you can try and check in web service message monitoring to see if there is any further information relating to error.
