We had worked with handling resources assigned both to source and destination locations.
This will assign a specific time window to each location to load/unload.
This way you force the system to send your vehicles through that specific transportation lane in the required schedule in order to meet those windows.
Other option would be to use schedules for your vehicle resources, but this way you will affect all the transportation lanes
In my experience, the first one (Handling Resources) involves less effort to manage on the ongoing.
