Is it possible to update the compensation portlet automatically based on employee seniority?
For example, we would need the portlet to add an amount or percentage increase to the base salary every 3 years.
As far as I know this is feasible in SAPR3, though, is it possible in Employee Central?

Actually, our situation is a little more complicated than that:
In Spain we have this scenario in which, by contract we have an annual gross salary agreed i.e. 30000€ =2500€ monthly gross
Salary tables are broken down into different components.
Base salary: 1000
Plus Convenio/ Bonus: 200
And then sometimes the employer agrees to reach the 2500 of gross amount, includes a component which is called “Mejora Vountaria/ Voluntary bonus” which is the result of the total 2500 gross monthly salary minus the total sum of salary components
2500-1200 = 1300 Mejora voluntaria
Likewise, a peculiarity that this pay component presents is that is absorbable which means that if the employee gets a salary raise because of his seniority, this component will decrease in the same amount of the increase. By way of illustration:
Every 2 years of seniority the employee has to receive 100 euros (let´s call it=seniority plus)
Therefore
1000 Base Salary
200 plus convenio
Now this new component “Seniority plus”=100
Then the voluntary bonus will be
2500-1000+200+100=1200€
in summary, what happens is that when the amount of the seniority pay component increases, the voluntary improvement component should automatically decrease by the same amount.
Then, what we need is that in compensation portlet appeared these components as of that date doing the correct calculations, and update it automatically.
We know that there is a functionality in which we can automate salaries according to seniority.

But, Is it possible to make the system recognize the total gross salary, deduct the rest of the components to know how much should the mejora voluntaria be reduced? All automatically without any manual changes?

There are 2 ways to my knowledge.
If salaries are determined based on pay scale levels and fixed salaries tied to those pay scale levels, you could automate it via Automatic Pay Scale Progression: Employee Central: Managing Pay Scale Progression | SAP Blogs.
If that is not the case, the best you could do is create an integration center job and schedule that.
