I want to customize the email notification template for enrollment, but just for a single item. I don"t want to change the global template. I want to do that inside one item , and in the "Manage Email Notification" tab. But my question is, will that trigger email automatically each time after the item is assigned to the user (like the global template works) or the admin need to do manually from "Email notification" tab of the item , each time.
Request clarification before answering.

The email trigger remains the same, just the email template (KBA 2274848) will come from a different place, as that was modified at the item level, in this case.
