How do I set the duration of a Highlight being on screen, then having one fade away and another show?

You mean the action Highlight in a Simulation? You should only ever see one at a time, as the macros are only played one at a time. They will display for as long as necessary (Display Duration in Demo mode, and until the Learner advances in Practiced). But see also the Playback Settings > General > Highlights > Type and Effect settings.

You can achieve this by using the Explanation bubble instead of the Highlight. The Explanation macro has a unique feature "Show Highlight in the Demo Mode". Once the Explanation bubble is closed (manually by the user or automatically due to the duration expiration) the highlight will also disappear.
