Hello, is the Disclosure Management version 10.1 supported for SQL Server 2016 with backward compatibilty mode set back to 2012?

No, it is not
The good news is - both Windows server 2016 and SQL Server 2016 support is scheduled for DM SP12Patch01 (Stack1201)
This is scheduled for release in June
