Can I create a purchase order using reference purchasing organization ?
Request clarification before answering.
Hi waman you can copy a purchasing organization in SPRO: Enterprise Structure -> Definition -> Materials Management -> Maintain purchasing organization.
However I don't know of a "define, copy, delete, check" transaction for purchasing organization, similar to "define, copy, delete, check" for plants or warehouses.

I cannot understand - why don't you try it in your system?
This is a lot faster and more useful for your personal development compared to asking a question here.
In short - whether you can create a PO for a specific purchasing organization or not depends on the configuration and on the master data, none of which are known to people with no access to your system.
