How to assign qualifications to the positions for the purpose of profile match up?
Request clarification before answering.
Use transaction PPPM (Change Profile). From there, search and select the position of interest. Then, assign a qualification (in the form of a requirement) against the position by clicking on the Paper (Create) icon. You can search for the qualification of interest, and select it. If you need to select a particular proficiency for the requirement (qual), you can define that as well.

I know this thread is ancient, but I still feel the need to point out that the answer that is marked as "correct" is in fact wrong, while what Julie said is correct, so she would have deserved the points. You cannot maintain the S <-> Q-relation in PO13, because the B 031-relation for object type Q is marked as "not directly maintainable" in SAP standard customizing. Of course you could change that setting, but that borders to a modification and carries comparable risks.
Qualifications for positions are being maintained through transaction PPPM instead.
